- Null mutant flies show a diminished change in lifespan in response to dietary restriction, which can be rescued upon ubiquitously expression of wild-type Thor (d4E-BP). Overexpression of wild type 4E-BP caused no change in lifespan of males or females on rich food, while overexpression of a weak activated allele extended mean lifespan of females 14% (no significant extension in male flies). Induction of a strong allele extended male lifespan 11% and female lifespan 22% on rich food. Under dietary restriction, there was no additional lifespan extension.
